We describe and demonstrate a new nanometer-scale broadband light source. It is based on the grating-coupled excitation of surface plasmon polaritons (SPPs) on the shaft of a sharp conical metal taper with a tip radius of few tens of nanometers. Far-field excitation of linear nanoslit gratings results in the resonant generation of SPPs traveling over more than 10 mum to the tip apex and converging to an intense radiative local light spot. Such nanofabricated tips are expected to find various applications in nanospectroscopy, overcoming problems with background illumination in apertureless microscopy. 相似文献

One of the most important steps in a productive viral infection is when the virus fuses to a cell membrane and delivers its genome into the cell cytosol. This dynamic event is mediated by interactions between specific virus envelope proteins with their cell-bound receptors. This process is exemplified by Moloney murine leukemia virus (Mo-MLV) where envelope protein interaction with its receptor, mCAT-1, leads to virus-cell membrane fusion and infection of cells. Here, fluorescent nanoparticles (NPs) were coated with Mo-MLV derived membranes (Mo-NPs) by extrusion. Electron microscopy and biochemical analysis showed tight association of the virus membranes and NPs. The coated NPs mimic native virus by binding and entering only cells expressing the virus receptor. Confocal microscopy revealed that the coated NPs were taken up into endocytic compartments containing receptor and were also seen associated with caveolin, a marker of caveolae. To demonstrate that the Mo-NPs could escape endosomes and deliver a protein cargo into the cell cytosol, beta-lactamase (betalac) was covalently coupled to the Mo-NP cores and incubated with cells. betalac activity was only detected in the cytosol of mCAT-1-expressing cells. This is the first time that virus proteins have been used to specifically target NPs to receptor-bearing cells as well as penetration into the cell cytosol. Extrusion provides a rapid, detergent-free method to couple virus membranes to NPs and should be readily applicable for many other virus and NP types. 相似文献

Exploration for geothermal resources is often challenging because there are no geophysical techniques that provide direct images of the parameters of interest, such as porosity, permeability and fluid content. Magnetotelluric (MT) and seismic tomography methods yield information about subsurface distribution of resistivity and seismic velocity on similar scales and resolution. The lack of a fundamental law linking the two parameters, however, has limited joint interpretation to a qualitative analysis. By using a statistical approach in which the resistivity and velocity models are investigated in the joint parameter space, we are able to identify regions of high correlation and map these classes (or structures) back onto the spatial domain. This technique, applied to a seismic tomography-MT profile in the area of the Groß Schönebeck geothermal site, allows us to identify a number of classes in accordance with the local geology. In particular, a high-velocity, low-resistivity class is interpreted as related to areas with thinner layers of evaporites; regions where these sedimentary layers are highly fractured may be of higher permeability. 相似文献

Abstract The design of an optical element profile with specified transmission function for a given incident wave is of fundamental concern in optical design. A well-known example is the design of an aspherical surface in order to realize a spherical phase-only transmission. Various wave-optical system design methods lead to transmission functions as a first step. Then, often the thin-element approximation is applied in a second step to obtain an element structure with the desired transmission. However, if the refraction at the optical interface cannot be neglected, the thin-element approximation is not valid. In this case, a higher version of the local plane-interface approximation can be used for the element structure design. An algorithm for this model is introduced and its characteristics are discussed for the example of a non-paraxial Gaussian-to-tophat beam shaping element. 相似文献

A new solid tungsten divertor for the fusion experiment ASDEX Upgrade is under construction at present. A new divertor tile design has been developed to improve the thermal performance of the current divertor made of tungsten coated fine grain graphite. Compared to thin tungsten coatings, divertor tiles made of massive tungsten allow to extend the operational range and to study the plasma material interaction of tungsten in more detail. The improved design for the solid tungsten divertor was tested on different full scale prototypes with a hydrogen ion beam. The influence of a possible material degradation due to thermal cracking or recrystallization can be studied. Furthermore, intensive Finite Element Method (FEM) numerical analysis with the respective test parameters has been performed. The elastic–plastic calculation was applied to analyze thermal stress and the observed elastic and plastic deformation during the heat loading. Additionally, the knowledge gained by the tests and especially by the numerical analysis has been used to optimize the shape of the divertor tiles and the accompanying divertor support structure.This paper discusses the main results of the high heat flux tests and their numerical simulations. In addition, results from some special structural mechanic analysis by means of FEM tools are presented. Finally, first results from the numerical lifecycle analysis of the current tungsten tiles will be reported. 相似文献

This paper describes the design, implementation, and operation of the Video Real-Time (VRT) diagnostic system of the ASDEX Upgrade plasma experiment and its integration with the ASDEX Upgrade Discharge Control System (DCS). Hot spots produced by heating systems erroneously or accidentally hitting the vessel walls, or from objects in the vessel reaching into the plasma outer border, show up as bright areas in the videos during and after the reaction. A system to prevent damage to the machine by allowing for intervention in a running discharge of the experiment was proposed and implemented. The VRT was implemented on a multi-core real-time Linux system. Up to 16 analog video channels (color and b/w) are acquired and multiple regions of interest (ROI) are processed on each video frame. Detected critical states can be used to initiate appropriate reactions – e.g. gracefully terminate the discharge. The system has been in routine operation since 2007. 相似文献

The past few decades have witnessed some of the most important and revolutionizing advances in the field of asymmetric catalysis. Chemists no longer rely solely on natural sources as the starting point of their synthetic strategy, as in chiral pool or auxiliary-based synthesis. Instead, naturally occurring chiral motifs are selected and, either unchanged or after modification, used in substoichiometric amounts as chiral catalysts or ligands. In this way, they effectively transfer their chirality to prochiral substrates, thereby rapidly amplifying and diversifying the arsenal of useful chiral building blocks available to the synthetic community. A long-standing goal in the pursuit of new catalytic systems is the discovery of general catalysts. Ideally, such catalytic systems should be capable of promoting a large number of enantioselective reactions, via multiple modes of activation, with good substrate tolerance and high stereoselectivity. In this Account, we describe the synthetic usefulness, efficiency, selectivity, and robustness of the diarylprolinol silyl ether system as the catalyst in various reactions of aldehydes. Based on the diarylprolinol silyl ether system, several studies on enamine-mediated transformations of saturated aldehydes have resulted in the introduction of different functionalities into the α-position of aldehydes in a highly stereoselective manner. This HOMO-activation concept was later extended to include α,β-unsaturated aldehydes, which after condensation with the aminocatalyst generate a dienamine species capable of undergoing stereoselective Diels-Alder-type reactions. As a result, the effective functionalization of the γ-position of the aldehyde is achieved. Recently, the activation principle was further developed to include 2,4-dienals, which form trienamine intermediates upon condensation with the aminocatalyst. The trienamines effectively react with carbon-centered dienophiles, forming aldehyde products having up to four contiguous stereocenters. Because of the concerted nature of the reaction and the efficient catalyst shielding of the β-position, the stereoinduction is achieved at the remote ε-position of the original aldehyde. Complementary to the enamine-mediated activations, α,β-unsaturated aldehydes can also be efficiently functionalized by applying the diarylprolinol silyl ether system via conjugate addition through iminium-ion-mediated processes, that is, LUMO-activation. In such reactions, the aminocatalyst not only effectively shields one of the enantiotopic faces of the enal, it also ensures excellent chemoselectivity, affording 1,4-adducts as the only products. Several different carbon and heteroatom nucleophiles can be added in a highly stereoselective fashion. The ability of the catalysts to participate in various enamine- and iminium-ion-mediated processes also makes them ideal for the sequential addition of nucleophiles and electrophiles in a cascade manner. These cascade reactions thereby afford access to products having at least two stereocenters. In the years to come, the diarylprolinol silyl ether catalysts will probably maintain their prominent position as general catalysts in the field of aminocatalysis. Moreover, recent efforts devoted to mechanistic studies might soon engender further advances with this versatile catalytic system, particularly in the areas of activation modes, catalyst loadings, and industrial applications. 相似文献
